Macroscopy and histology assessments are essential tools for evaluating the pharmacodynamics of your product candidate on target organs and tissues. Macroscopy and histology assessments are performed by blinded experts using validated and published semi-quantitative scoring tables.

Macroscopy

Visual examination of tissue/ organ specimens by describing the size, the shape, the color and texture. This analysis is key to identify features that can guide further microscopic analysis.

  • Macroscopic evaluation of joint lesions according to the OARSI scoring system (synovium, cartilage, and bone sub-units)
  • Macroscopic evaluation of the synovial fluid
  • Biomechanics (MACH-1)

Histology

Examination of tissue/organ specimens at microscopic level to understand their structure, composition, and properties. This analysis is key to determine or verify the internal structure and morphology of samples, which can reveal important information about their function and about the efficacy of the product.

  • Histological processing of various hard and soft tissues (paraffin samples, cryosections)
  • Large panel of histological stainings (e.g. hematoxylin and eosin, safranin-O fast green, sirius red, toluidine blue, masson-goldner)
  • Histological evaluation of joint lesions according to OARSI and ICRS scoring grids
  • Immunostaining and immunohistochemistry with 22 validated targets specific to cartilage metabolism, muscle fibers and inflammation (specific target on demand)
  • Histomorphometry and quantification (on demand, e.g. cartilage proteoglycan content, bone content and architecture tissue cellularity)
